PhosphoIipid/Ca2+ -dependent protein kinase C (PKC) and oxytocin receptor were measured in sheep endometrial explants after culture for up
to 96 h. Oxytocin receptor binding and PKC activity were reduced by up to 90% in explants exposed to recombinant ovine trophoblast
interferon (rolFN-τ), recombinant bovine IFN-α1 or ovine conceptus secretory proteins (a source of IFN-τ). Inhibition occurred in both caruncular and intercaruncular endometrium
taken between days 7 and 10 of the oestrous cycle and in intercaruncular (but not caruncular) endometrium on day 6. Down-regulation
of PKC by continued exposure of expiants to 4β-phorbol myristate acetate, or treatment with PKC inhibitors reduced both oxytocin
receptor binding and PKC activity by up to 70%. Tyrosine kinase inhibitors were ineffective. Addition of oxytocin or progesterone,
which reduce oxytocin receptor bindingin vivo, also lowered oxytocin receptor bindingin vitro in the absence of any effect on PKC. The data indicate that IFN-τ inhibits oxytocin receptor synthesis by a mechanism involving
PKC inhibition, but that a non-PKC pathway also operates to control oxytocin receptor binding in non-pregnant animals. These
conclusions were supported by measuring PKC activity and oxytocin receptor binding in endometrium without culture. Prolonged
exposure of the endometrium to IFN-τin vivo may lead to PKC down regulation by a mechanism analogous to that involved in the action of continuous activation by agonist,
and this may represent one function of the prolonged secretion of IFN-τ over a 10-day period in early pregnancy. 相似文献

目的研究自制无创面罩垫对面型瘦小患者无创正压通气的改善作用及预后效果影响。方法选取2016年2月至2018年2月于佛山市第五人民医院接受无创正压通气治疗的面型瘦小患者80例,以随机抽签法分成实验组与对照组,各40例。对照组予以传统呼吸机面罩行无创正压通气,实验组在对照组的基础上加用自制无创面罩垫行无创正压通气。比较两组治疗前后血气分析指标水平、舒适度情况、面罩漏气、破损情况、压疮发生率、满意度情况。结果治疗后实验组PaO2水平显著高于对照组,而PaCO2水平显著低于对照组,实验组患者的舒适度显著高于对照组,实验组患者面罩漏气、破损情况及压疮发生率均显著低于对照组,实验组满意度显著高于对照组,差异均有统计学意义(均P<0.05)。结论自制无创面罩垫应用于无创正压通气治疗的面型瘦小患者中效果明显,有利于改善患者的血气分析状况,缓解传统呼吸机面罩所造成的不适症状,同时有利于降低面罩漏气、破损以及压疮发生风险,提高满意度。 相似文献

目的:探讨生长激素(growth hormone,GH)在体外受精-胚胎移植(in vitro fertilization-em-bryo transfer,IVF-ET)治疗中对子宫内膜发育不良者子宫内膜及临床结局的影响。方法:IVF-ET患者共67例,均由于子宫内膜发育不良而致IVF-ET种植失败,再次行IVF-ET治疗时随机分为研究组(加用GH治疗,32例)和对照组(未使用GH治疗,35例),统计分析患者的临床结局。结果:患者Gn用量、平均获卵数、受精率、优质胚胎率组间无统计学差异(P>0.05);研究组子宫内膜厚度、形态及子宫内膜和内膜下血流均较对照组有明显改善(P<0.01);研究组的胚胎种植率(31.4%)及临床妊娠率(39.5%)明显高于对照组(15.7%和19.8%)(P<0.01),早期流产率明显减低(14.6%vs 21.4%)(P<0.01)。结论:对内膜发育不良的患者进行IVF-ET治疗时加用GH可能是通过改善子宫内膜对胚胎的容受性,从而提高胚胎种植率及临床妊娠率,降低早期流产率。 相似文献

目的探讨超声引导下子宫内膜消融术在良性疾病导致的异常子宫出血中的治疗效果。方法 应用自凝刀对114例患者在超声监护下行子宫内膜射频消融术,术后定期随访,观察疗效。结果自凝刀子宫内膜消融术治疗子宫出血平均(12.32±4.51)min,96例治愈,18例有效,总有效率100%,无严重并发症。结论超声引导下子宫内膜消融术治疗子宫异常出血是一种安全、疗效好、简单、快速的治疗方法。 相似文献

MicroRNAs (miRNAs) are a recently discovered class of non-coding RNAs that are expressed in many cell types, where they regulate the expression of complementary RNAs, thus modulating the stability and translation of mRNAs, miRNAs are predicted to regulate the expression of -50% of all protein coding genes in mammals. Therefore, they participate in virtually all cellular processes investigated so far. Altered miRNAs expressions are associated with both physiological (pregnancy) and pathological processes (cancer). As the dynamic maternal-fetal interface plays a critical role in the maintenance of successful pregnancy, it is not surprising that the miRNAs that are unique to reproductive tissues are abundantly expressed. Research in this field has demonstrated the presence and dysregulation of a distinct set of pregnancy-associated miRNAs; however, most studies have centered on localizing various miRNAs in reproductive microdomains associated with normal or complicated pregnancies. Although several independent miRNA regulatory mechanisms associated with endometrial receptivity, immune cells, angiogenesis and placental development have been studied, miRNA-mediated regulation of pregnancy remains poorly understood. This review provides a summary of the current data on miRNA regulation as well as functional profiles of miRNAs that are found in the uterus, in immune cells associated with maternal tolerance to the fetus, and those involved in angiogenesis and placental development. 相似文献
